Notes Angiopoietins are members of the vascular endothelial growth factor family and the only known growth factors largely specific for vascular endothelium. Angiopoietin-1, angiopoietin-2, and angiopoietin-4 participate in the formation of blood vessels. Angiopoietin-like 3 is predominantly expressed in the liver, however, its exact function is not known. The ANGPTL3 protein has the characteristic structure of angiopoietins: a signal peptide, an extended helical domain predicted to form dimeric or trimeric coiled-coils, a short linker peptide, and a globular fibrinogen homology domain (FHD). ANGPTL3 also does not contain the characteristic calcium-binding motif found in other angiopoietins. ANGPTL3 stimulates endothelial cell adhesion and migration via integrin alpha vbeta 3 and induces blood vessel formation in vivo. Angptl3 gene is a direct target of the liver X receptor (LXR). Data show that angiopoietin-like protein 3 (ANGPTL3) targets adipose cells and induces lipolysis. The cleavage of ANGPTL3 at two sites is important for the activation of ANGPTL3 in vivo.
